  Occurrence. Pancreatic spasmolytic peptide (PSP) [82987-77-7], Mr 11 711, is a pyroglutamyl peptide containing 106 amino acids and seven disulfide bridges [2018]. Immunoreactive PSP is found in low concentrations in many organs, but substantial amounts only occur in the pancreas. There is about 50 % sequence homology between p-PSP-(14 – 49) and p-PSP-(63 – 98). There is great similarity between the sequences of p-PSP and the protein pS2 which was isolated from human gastric juice [2019] and is induced by estrogen in the human breast cancer cell line MCF-7 [2020]. pS2 is identical to the human epidermal growth factor-like immunoreactive factor [2021]. A protein from the skin gland of Xenopus laevis, known as the APEG protein [2022], has a sequence at its C-terminus that is very similar to p-PSP and h-pS2. Another protein from Xenopus laevis, which was characterized via the cDNA structure, contains, after possible processing, two basic sequences (spasmolysin I and spasmolysin II) that are very similar to PSP [2023].

  Biological Effects. p-PSP inhibits the activity of adenylate cyclase in the intestinal mucosal cell membranes of the rat [2024]. It inhibits gastrointestinal motility and the secretion of gastric acid in laboratory animals after parenteral and oral application. It also reduces the pressure and the wave frequency of the Oddi sphincter [2025]. PSP acts like a growth factor on MCF-7 cells [2019].
